Qualified participants must:
- Have a BMI 35 kg/m² or higher
- Have tried to lose weight with diet and exercise at least once in the past 6 months without lasting success
Answers Must Be No
- Have you taken any of these medications within the past 3 months?
- Semaglutide (Ozempic, Wegovy, Rybelsus)
- Tirzepatide (Mounjaro, Zepbound)
- Liraglutide 3 mg (Saxenda)
- Any other GLP-1 receptor agonist medication (injectable medications for diabetes or weight loss)
- Any other prescription weight loss medications like orlistat, phentermine, lorcaserin, naltrexone/bupropion combination, or phentermine/topiramate combination
- Have you ever had, or are you planning to have, any weight loss surgery or procedures such as:
- Gastric bypass, Gastric sleeve, Lap band, Gastric balloon
- Duodenal-jejunal liner, Gastric artery embolization
- Any endoscopic weight loss procedure
- (Note: Liposuction is okay if it was more than 1 year ago. If you had a device removed, it must have been more than 6 months ago.)
- Have you ever been diagnosed with diabetes (either Type 1 or Type 2) or have an A1c ≥6.5%?
- Are you currently taking any medications for blood sugar control, such as: Insulin (any type), Metformin (Glucophage), Sulfonylureas like glipizide (Glucotrol) or glyburide (Diabeta) or any other diabetes medications?
- Has your weight changed by more than 5% (up or down) in the past 3 months? (For example, if you weigh 200 pounds, that would be a 10-pound change.)
- Have you started taking any of these types of medications within the past 3 months?
- Tricyclic antidepressants (like imipramine or amitriptyline)
- Atypical antipsychotics (like clozapine, olanzapine, quetiapine)
- Mood stabilizers (like valproic acid, lithium)
- Mirtazapine (Remeron)
- Paroxetine (Paxil)
- Have you taken, or are you likely to need, daily steroid pills or injections for more than 2 weeks within the past 3 months or in the next 12 months for significant active autoimmune conditions like lupus or rheumatoid arthritis? (Inhalers, nasal sprays, eye drops, skin creams, and joint injections are okay.)
- Are you allergic or have you had a bad reaction to any GLP-1 medications (like Ozempic, Wegovy, Trulicity, Victoza, Saxenda, Mounjaro, or Zepbound)?
- Within the past 2 years, have you had active or unstable (have you started any new meds or had any changes in dose) major depression, schizophrenia, bipolar disorder, or another serious mood or anxiety disorder?
- Have you been diagnosed with an eating disorder such as anorexia nervosa or bulimia nervosa?
- Do you have any of the following heart conditions:
- Nonischemic cardiomyopathy (a type of heart muscle disease not caused by blocked arteries)
- Class IV heart failure (the most severe type where you have symptoms even at rest)
- Significant irregular heart rhythms
- Have you had cancer within the past 5 years? (Exceptions: Basal or squamous cell skin cancer, cervical carcinoma in situ, or prostate cancer in situ are okay.)
Answers Must Be Yes:
- Are you willing to complete a self-injection training during the screening visit, including injecting your abdomen with saline under staff supervision?
- Are you willing to come to the clinic every 4 weeks for about a year and a half?
- Are you willing and able to follow a reduced-calorie diet and mild-to-moderate exercise program throughout the study?
- If female of childbearing potential and sexually active, do you agree to use 2 highly effective forms of contraception?
- Male participants: Do you agree to refrain from donating sperm during the study and for at least 35 days after the end of the study?
